Six new Phytophthora species from ITS Clade 7a including two sexually functional heterothallic hybrid species detected in natural ecosystems in Taiwan. Naturalis
Jung, T.; Jung, M.H.; Scanu, B.; Seress, D.; Kovács, G.M.; Maia, C.; Pérez-Sierra, A.; Chang, T.-T.; Chandelier, A.; Heungens, K.; Poucke, K. van; Abad-Campos, P.; Léon, M.; Cacciola, S.O.; Bakonyi, J..
During a survey of Phytophthora diversity in natural ecosystems in Taiwan six new species were detected. Multigene phylogeny based on the nuclear ITS, ß-tubulin and HSP90 and the mitochondrial cox1 and NADH1 gene sequences demonstrated that they belong to ITS Clade 7a with P. europaea, P. uniformis, P. rubi and P. cambivora being their closest relatives. All six new species differed from each other and from related species by a unique combination of morphological characters, the breeding system, cardinal temperatures and growth rates. Four homothallic species, P. attenuata, P. flexuosa, P. formosa and P. intricata, were isolated from rhizosphere soil of healthy forests of Fagus hayatae, Quercus glandulifera, Q. tarokoensis, Castanopsis carlesii,...

Spatial and temporal extension of eutrophication associated with shrimp farm wastewater discharges in the New Caledonia lagoon ArchiMer
Thomas, Yoann; Courties, Claude; El Helwe, Yasmin; Herbland, Alain; Lemonnier, Hugues.
Shrimp farming in New Caledonia typically uses a flow-through system with water exchange rates as a tool to maintain optimum hydrological and biological parameters for the crop. Moreover, the effluent shows hydrobiological characteristics (minerals, phytoplankton biomass and organic matter) significantly higher than that of the receiving environment. Separate surveys were carried out in a bay (CH Bay) with a medium-size intensive farm (30 ha) (PO) and in a mangrove-lined creek (TE Creek) near a larger semi-intensive farm (133 ha) (SO). Net loads of nitrogen exported from the semi-intensive farm and the intensive farm amounted to 0.68 and 1.36 kg ha(-1) day(-1), respectively. At CH Bay, discharge effects were spatially limited and clearly restricted to...

Spatial and temporal variability of the pico- and nanophytoplankton and bacterioplankton in a deep Polynesian atoll lagoon ArchiMer
Thomas, Y.; Garen, Pierre; Courties, C.; Charpy, L..
Spatial and temporal scales of variability in phyto- and bacterioplankton abundance were investigated in a deep atoll lagoon (Ahe, French Polynesia), Ahe Lagoon showed a classical phytoplankton composition, but its picophytoplankton (Prochlorococcus, Synechococcus and picoeukaryotes) concentrations are higher than the mean for Polynesian atoll lagoons. Phyto- and bacterioplankton descriptors were highly variable, with mean CVs of >20% in most cases: day night, day-to-day, vertical, lagoonal and seasonal. Phytoplankton biomass, which was estimated by in vivo fluorescence, was higher in the deep layer than in the upper layer. This pattern was detected on a day night scale, showing transitory vertical gradients, and through long-term events that were...

Study of atrazine effects on Pacific oyster, Crassostrea gigas, haemocytes ArchiMer
Gagnaire, Beatrice; Renault, Tristan; Bouilly, Karine; Lapegue, Sylvie; Thomas-guyon, H.
Shellfish farming is an important economic activity around the world. This activity often takes place in areas subjected to various recurring pollutions. The recrudescent use of herbicides in agriculture including atrazine implies pollutant transfer towards aquatic environment in estuarine areas. Harmful effects of such substances on animals in marine environment, particularly on cultured bivalves, are poorly documented. Bivalve molluscs such as mussels and oysters have been postulated as ideal indicator organisms because of their way of life. They filter large volumes of seawater and may therefore accumulate and concentrate eontaJ.11inants within their tissues. Moreover, development of techniques allowing effect analysis of such compounds on bivalve...

Survey for neoplasia in Macoma balthica from the Gulf of Gdansk by flow cytometry ArchiMer
Smolarz, Kasia; Renault, Tristan; Soletchnik, Patrick; Wolowicz, Macciek.
Using flow cytometry, 234 Macoma balthica were examined during a survey to determine frequency of neoplasia in the Gulf of Gdansk (Poland). Clams were collected in 4 locations and DNA content in gill tissue cells was determined by flow cytometry using propidium iodide staining. Cell permeabilization was induced by osmotic shock. Prevalence of neoplasia ranged from 9.6 to 26.7 % depending on location. DNA content in aneuploid cells was higher than in normal dividing cells. The fluorescence value for aneuploid cells corresponded to tetraploid/pentaploid cells. Three stages of neoplasia were defined, based on the percentage of aneuploid cells determined by flow cytometry. Histopathological and cytogenetic analyses were also carried out on the same clams for...

TOPHYPAC. Tolérance des communautés phytoplanctoniques aux phytosanitaires dans le panache de la Charente. Programme Évaluation et réduction des risques liés à l’utilisation des Pesticides ArchiMer
Stachowski-haberkorn, Sabine; Guesdon, Stephane; Bechemin, Christian; Chiffoleau, Jean-francois; Brach-papa, Christophe; Soudant, Philippe; Beker, Beatriz; Jadas-hécart, Alain.
TOPHYPAC is a field study that was conducted in the Charente river estuary (Marennes-Oléron bay). The main goal was to assess the possible impact of chemical contamination with copper and pesticides on phytoplankton communities. Physical, chemical (nutrients, metals and pesticides) and biological (phytoplankton community) parameters were monitored in the field during three years, from 2011 to 2014. In 2012 and 2013, field sampling was coupled with experimental exposure of phytoplankton communities to copper and to an herbicide cocktail, after transportation to the lab: the effects were assessed using photosynthesis endpoint and cellular densities. TOPHYPAC allowed to set the framework of Charente estuary contamination with pesticides: herbicides glyphosate...

Trophic relationships between the farmed pearl oyster Pinctada margaritifera and its epibionts revealed by stable isotopes and feeding experiments ArchiMer
Lacoste, Elise; Raimbault, Patrick; Harmelin-vivien, Mireille; Gaertner-mazouni, Nabila.
Bivalve cultures support a host of epibionts, mainly suspension feeders, which can compete for food resources with the cultivated bivalves. However, the magnitude of interspecific competition for food in bivalve aquaculture settings remains inconclusive, especially in tropical areas. We investigated the interactions for food between the farmed pearl oyster Pinctada margaritifera and its epibionts, using stable isotope analysis and feeding experiments. Inter-and intraspecific variations of delta C-13 and delta N-15 stable isotope ratios (SIRs) were determined for oysters in the presence or absence of epibionts. The diet of the most abundant epibionts, Herdmania momus and Didemnum sp., was specified using isotope measurements and flow cytometry during...

Trypan blue exclusion assay by flow cytometry BJMBR
Avelar-Freitas,B.A.; Almeida,V.G.; Pinto,M.C.X.; Mourão,F.A.G.; Massensini,A.R.; Martins-Filho,O.A.; Rocha-Vieira,E.; Brito-Melo,G.E.A..
Dye exclusion tests are used to determine the number of live and dead cells. These assays are based on the principle that intact plasma membranes in live cells exclude specific dyes, whereas dead cells do not. Although widely used, the trypan blue (TB) exclusion assay has limitations. The dye can be incorporated by live cells after a short exposure time, and personal reliability, related to the expertise of the analyst, can affect the results. We propose an alternative assay for evaluating cell viability that combines the TB exclusion test and the high sensitivity of the flow cytometry technique. Previous studies have demonstrated the ability of TB to emit fluorescence when complexed with proteins. According to our results, TB/bovine serum albumin and...

Escandón,Alejandro S; Miyajima,Ikuo; Alderete,Marisol; Hagiwara,Juan Carlos; Facciuto,Gabriela; Mata,Diego; Soto,Silvina M.
The genus Scoparia is native from Argentina. The diversity of colours and shape of their flowers bestows this genus a very interesting ornamental potential. The purpose of the present study is to explore the Scoparia species germplasm by means of in vitro polyploidization in order to improve their ornamental qualities. Accessions of S. montevidiensis var. montevidiensis, S. montevidiensis var. glandulifera, S. nudicaulis, S. hasleriana and S. dulcis were collected and maintained under greenhouse conditions. The Murashige-Skoog medium, supplemented with 0.25 mg/L BAP was used for the nodal segments multiplication of the five Scoparia species. Except for S. hasleriana, the multiplication rate of the other species ranged between 10 and 12 shoots per explant....
